SNAP-8 (Acetyl Octapeptide-3)
SNAP-8 is a longer, eight-amino-acid version of Argireline for cosmetics. It is studied as an ingredient that relaxes the facial expression muscles more strongly and reduces wrinkles at a lower concentration.
Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH₂ — 8 aa (extended Argireline + Ala-Asp). A more complete mimicry of the N-terminal domain of SNAP-25.
A second-generation muscle relaxant — an extended version of Argireline. According to the manufacturer: tighter binding → a more pronounced effect at lower concentrations. −38% wrinkles at 3% over 28 days. Microneedle patches (0.03%): improved transdermal delivery.
Competitively inhibits assembly of the SNARE complex (SNAP-25+Syntaxin+VAMP) → ↓ACh exocytosis. 8 aa = tighter binding to the target vs the 6 aa of Argireline. Result: a more pronounced ↓contraction of facial expression muscles.
Studies used 3–5% in a serum twice a day. Microneedle patches 0.03% — 24 volunteers, 28 days. Clinical result: −38% at 3% over 28 days.
Level B Human observational data. There are studies in humans, but they are small, uncontrolled, or cohort studies. The data are encouraging but need confirmation. Clinical studies in formulations. Standalone placebo-controlled RCTs are limited.
⚠️ Cosmetic ingredient (Lipotec/Lubrizol). Fewer standalone RCTs than for Argireline.
