Matrixyl (Palmitoyl Pentapeptide-4 / pal-KTTKS)
Matrixyl is one of the most studied cosmetic peptides — a small piece of the collagen precursor molecule. It is studied as an ingredient that nudges the skin to produce new collagen and makes wrinkles shallower.
Pal-Lys-Thr-Thr-Lys-Ser — 5 aa + a palmitoyl group. A fragment of the C-terminal propeptide of procollagen I. Patented (Sederma).
A collagen synthesis stimulator — one of the most studied cosmetic peptides. Double-blind RCT 2023 + landmark clinical study 2005. Significant reduction in wrinkle depth and volume after 8–16 weeks. Mechanism: mimics collagen breakdown products → activates synthesis of new collagen.
KTTKS = a matrikine fragment of procollagen I → fibroblast receptors → activates the TGF-β cascade → ↑synthesis of collagen I, III, IV + fibronectin + hyaluronic acid. The palmitoyl group provides transdermal delivery across the skin's lipid barrier.
Studies used 2–5 ppm in a serum or cream twice a day for an 8–16-week course. A lasting effect was noted with regular use.
Level A Human clinical trials. There are randomized controlled trials (RCTs), or the compound has been approved by a regulatory agency. The most reliable level. Double-blind RCT 2023 + a solid clinical evidence base.
⚠️ Patented ingredient (Sederma/Croda). Cosmetic, not pharmaceutical.
Research peptides are laboratory reagents (Research Use Only). They are not medicines and are not intended for use in humans or animals.
